What's the difference between 'large amount' and 'large number' ? And when to use it? :)

We use "amount" for things that can't be counted (or mass nouns). We use "number" for things that can be counted.
We drink large AMOUNTS of MILK every morning.
The teacher gives a large AMOUNT of HOMEWORK every Friday.
I see a large NUMBER of PEOPLE at the concert.
A large NUMBER of CRAYONS are missing from the box.

That's a tricky word. Since "qua" means "as being," a sentence such as this would be correct:
High fashion qua fashion appeals to a very small group of people.
The movie critic becomes upset whenever he's asked to discuss slasher films qua films.

Maybe forgive someone is easier .. But forget what had been done to ourself seriously difficult .. Is the grammar in this sentences are true ?

what does off of mean? and is it any different from off?

"Off" can be the opposite of "on," but "off of" can't: He turned the TV off.

what's the meaning of '50shades' ?

"50 Shades of Grey" is the title of a popular book that will be turned into a movie. In the book, the protagonist, Christian Grey, calls himself "50 shades of fu*ked up, hence the name "50 Shades of Grey."
The number 50 is important only because that's what Christian Grey picked to describe himself. There is no other significance of that number. In other words, the book could have just as easily been called "38 Shades of Grey" (although that doesn't sound as good).
By the way, we have not read the book, nor do we have any desire to do so. Apparently, it's nothing more than bondage porn.

"I am going to read those books as soon as I have some free time" is the best way to express that idea.
"Waiting list" doesn't work because a "waiting list" is for people who are waiting for housing or admission to a school.

I am stressed or I am stressful.

"I am stressed" = I am worried, anxious, nervous, etc.
"I am stressful" = I cause stress to others.
In other words, we generally use "stressful" to describe things such as exams, trials, obstacles, situations, etc.

"It's evening already?" and "It's already evening?" are right.
They're comparable to "It's noon already?" and "It's already noon?"

When to use ain't? Is it singular or plural?

Both.
I ain't lying.
We ain't going there.
AIN'T is nonstandard and extremely informal. Use it only with friends; never use it in formal writing.

Which is true; "what is the difference between..." or "what are the difference between..."

Because "difference" is singular, "what IS the difference between ..." is correct.

With "weekend," we usually use "over":
What will you be doing over the weekend?
We can also just go with "What will you be doing this weekend?"

With "remark," we usually use "make" or "made" (depending on the tense of the sentence):
He made an insensitive remark about cats.
Don't make offensive remarks.

How to use thw word remark. Is it a verb or a noun

"Remark" is both a noun and a verb.
"The room is too cold," remarked Jo. (verb)
Jo's remark hurt my feelings. (noun)
